Simple, straightforward and fast sample preparation of total daidzein, genistein and equol analysis in urine

Isoflavones and isoflavandiols have potential protective effects against a number of complex diseases. However, in order to investigate the health benefits of these phytochemicals, it is necessary to develop simple, fast and reliable analytical methods for their quantification in biological fluids without the use of extraction or the preconcentration techniques inherent in existing methods. The current study describes a simple and fast method for the preparation of human urine samples for LC-MS/MS analysis of daidzein and genistein (isoflavones) and equol (isoflavandiol). Sample preparation includes addition only of dimethylformamide (DMF) and formic acid (FA) after complete enzymatic hydrolysis of their metabolites by a ß-glucuronidase and sulfatase mixture. Recoveries were >90%, and the relative standard deviation for intra- day (7%) and inter-day (10 days) <15% were good. This method can be used for carrying out studies on the bioavailability or cross-sectional studies of isoflavones in urines.
